Indication for using Baricinix-4mg(Baricitinib):

Baricitinib is a prescription medication used to treat the following specific conditions:

Rheumatoid arthritis (RA) in adults who have not responded well to one or more tumor necrosis factor (TNF) inhibitor medications.

Alopecia Areata (AA) in adults with severe AA is defined as a Scalp Hair Loss Area Classification (SHLAC) score of ≥50%.

Hospitalized adults with COVID-19 who require supplemental oxygen, a ventilator, or who need extracorporeal membrane oxygenation (ECMO).

Baricinix-4mg(Baricitinib) is a disease-modifying antirheumatic drug (DMARD) approved by the FDA for treating moderate to severely active rheumatoid arthritis in adults who have not responded well to other DMARDs. Clinical trials have shown significant improvement and inhibition of disease progression compared to placebo. In 2020, the FDA issued an emergency use authorization for baricitinib in combination with remdesivir for COVID-19 treatment in hospitalized patients. Baricitinib is not FDA-approved as a standalone treatment for COVID-19. Mechanism of Action of  Baricinix-4mg(Baricitinib):

• Baricinix-4mg(Baricitinib) is a Janus kinase (JAK) inhibitor that works by blocking the activity of JAKs, which can reduce inflammation and improve symptoms.
• It inhibits the activity of JAK1 and JAK2, two of the four JAK proteins involved in the signaling pathways of pro-inflammatory cytokines. By inhibiting JAK1 and JAK2, baricitinib can block the signaling of these cytokines and reduce inflammation.
• It also has other anti-inflammatory effects, such as reducing the production of inflammatory mediators and suppressing immune cell activation. Baricitinib has a higher affinity for JAK1 and JAK2, inhibiting JAK proteins, preventing phosphorylation and activation of STATs, and modulating the signaling pathway of various interleukins, interferons, and growth factors. It also decreases the proliferation of JAK1/JAK2 expression in mutated cells and induces cell apoptosis.

Dosage and Administration:

Rheumatoid arthritis

The recommended dosage of baricitinib for rheumatoid arthritis is 2 mg once daily orally, with or without food.

Alopecia areata

The recommended dosage of baricitinib for alopecia areata is 2 mg once daily orally, with or without food. The dosage may be increased to 4 mg once daily if the response to treatment is not adequate.

COVID-19

The recommended dosage of baricitinib for COVID-19 is 4 mg once daily orally, with or without food, for 14 days or until hospital discharge, whichever occurs first.

Dosage adjustments

Dosage adjustments may be necessary for patients with certain conditions, such as kidney disease or liver disease. Dosage adjustments may also be necessary for patients who are taking certain medications that can interact with baricitinib.

Administration

Baricitinib is taken orally once daily. It is administrable with or without food.
